Non-opioid analgesics form reversible hydrogen bonds near the active centrum of cyclooxygenase (COX)-1. This prevents (1) aspirin entrance to the hydrophobic channel; (2) irreversible acetylation of Ser530; and (3) platelet inhibition for the remainder of the platelets life-span.
