Figure 1 Protective effects of Ab-DeliverIN (DelIN)-mediated intracellular delivery of polyclonal anti-deglycosylated ricin A-chain antibody (anti-dgA Ab) (A) and monoclonal RAC18 anti-ricin A-chain monoclonal antibody (RAC18 mAb) (B) against ricin-induced cytotoxicity.
RAW264.7 cells were incubated with naked antibodies (dgA Ab, RAC18 mAb or control IgG) (no liposome encapsulation) or DelIN-encapsulated antibodies (DelIN-dgA Ab, DelI-RAC18 mAb or DelIN-control IgG) for 8 h. After 3 washes with PBS to remove extracellular antibodies, ricin (100 ng/mL) was added and incubated for additional 24 h. Cell viability was quantitated using the MTT assay. As controls for the neutralizing effects of anti-ricin antibodies, ricin was co-incubated with naked dgA Ab, RAC18 mAb or control IgG.
